The present invention relates to a flavoprotein improved LOV (iLOV) variant with increased fluorescence intensity compared to iLOV. In addition, the present invention relates to a method for screening an iLOV variant. The iLOV variant of the present invention has increased fluorescence intensity and quantum yield compared to existing LOV proteins or iLOV proteins, such that the iLOV variant can be utilized to confirm the expression of a target protein or to separate and purify the expressed target protein regardless of whether or not oxygen is present.

La présente invention concerne un variant de flavoprotéine à domaine LOV amélioré (iLOV) présentant une intensité de fluorescence accrue par rapport à iLOV. De plus, la présente invention concerne un procédé de criblage d'un variant d'iLOV. Le variant d'iLOV de la présente invention présente une intensité de fluorescence et un rendement quantique accrus par comparaison avec des protéines LOV ou iLOV existantes, de telle sorte que le variant d'iLOV peut être utilisé pour confirmer l'expression d'une protéine cible ou pour séparer et purifier la protéine cible exprimée, indépendamment de la présence ou pas d'oxygène.

본 발명은 iLOV(flavoprotein improved LOV) 대비 형광세기가 증가된 iLOV 변이체에 관한 것이다. 또한, 본 발명은 iLOV 변이체의 스크리닝 방법에 관한 것이다. 본 발명의 iLOV 변이체는 기존의 LOV 단백질 또는 iLOV 단백질 보다 형광세기 및 양자수율이 증가되어 산소의 존재 유무에 상관없이 목적 단백질의 발현 여부를 확인하거나 발현된 목적 단백질을 분리정제하는데 유용하게 활용할 수 있다.
