Abstract
(EN) The present invention provides a comprehensive assay capable of comprehensively measuring interactors for various different types of proteins. The present invention provides a system for detecting the activation of a protein by integrating changes in the transcription regulatory region of a gene the expression of which is changed by activation of the protein via a plurality of factors. The stimulation response ratio of transcription activity before and after stimulation by ligands of each transcription regulatory region is synergistically integrated to permit construction of a highly sensitive reporter assay system.
(FR) La présente invention concerne une méthode d'analyse intégrée apte à mesurer de manière intégrée des interacteurs pour différents types de protéines variées. La présente invention concerne un système permettant de détecter l'activation d'une protéine en intégrant les changements de la région régulatrice de la transcription d'un gène dont l'expression est modifiée par l'activation de la protéine par l'intermédiaire d'une multiplicité de facteurs. Le rapport de la réponse de stimulation de l'activité de transcription avant et après la stimulation par des ligands de chaque région régulatrice de la transcription est intégré de manière synergique pour permettre la construction d'un système analytique rapporteur hautement sensible.
(JA) 本発明は、タンパク質の種々の異なる種類に対する相互作用因子を網羅的に測定することができる網羅的なアッセイを提供する。 本発明では、タンパク質の活性化によって発現が変化する遺伝子の転写調節領域における変化を複数の因子を介在させて統合することによって、当該タンパク質の活性化を検出する系が提供される。それぞれの転写調節領域が有するリガンド刺激前後での転写活性の刺激応答比が相乗的に統合され、高感度なレポーターアッセイ系が構築され得る。
