(JA) 細胞培養基材、がん細胞集合体及び該基材を用いたその製造方法、並びに該がん細胞集合体を用いた薬剤のスクリーニング方法
Abstract:
(EN) [Problem] An objective of the present invention is the in-vitro production of a cancer cell population retaining native biological properties, such as morphological polarity and tissue motion polarity, of the original cancer tissue. [Solution] The present invention pertains to a cell culture substrate comprising a base material and a biocompatible polymer layer, the cell culture substrate having, at prescribed intervals on the substrate surface, a plurality of rough sections having a prescribed surface structure having a prescribed shape and not covered by the biocompatibile polymer layer. The present invention makes it possible to use a very simple process, namely culturing cancer cells on a cell culture substrate having a prescribed structure, to obtain, in a living state, a cancer cell aggregate having the same morphological polarity and tissue motion polarity as is observed in-vivo, whereby the heretofore impossible live imaging of micro tumors in-vitro is made possible. Since it is considered that said cancer cell aggregate would reproduce the in-vivo sequence of cancer development, proliferation, invasion, metastasis and recurrence, the cancer cell aggregate could be used as a research tool for cancer research or for screening cancer drugs.
(FR) Un objectif de la présente invention concerne la production in vitro d'une population de cellules cancéreuses conservant des propriétés biologiques natives, telles que la polarité morphologique et la polarité de mouvement du tissu, du tissu cancéreux d'origine. La solution selon l'invention porte sur un substrat de culture cellulaire comprenant un matériau de base et une couche polymère biocompatible, le substrat de culture cellulaire ayant, à des intervalles prescrits sur la surface du substrat, une pluralité de sections rugueuses ayant une structure de surface prescrite ayant une forme prescrite et non couverte par la couche polymère biocompatible. La présente invention permet d'utiliser un procédé très simple, à savoir la culture de cellules cancéreuses sur un substrat de culture cellulaire ayant une structure prescrite, pour obtenir, sous un état vivant, un agrégat de cellules cancéreuses ayant la même polarité morphologique et polarité de mouvement du tissu que celles observées in vivo, moyennant quoi l'imagerie en direct jusqu'ici impossible des micro-tumeurs in vitro est rendue possible. Dans la mesure où il est considéré que ledit agrégat de cellules cancéreuses reproduirait la séquence in vivo du développement, de la prolifération, de l'invasion, des métastases et de la récurrence du cancer, l'agrégat de cellules cancéreuses pourrait être utilisé comme outil de recherche pour la recherche sur le cancer ou pour le criblage de médicaments contre le cancer.
(JA) 【課題】 本発明は、形態学的極性や組織運動極性といったがん組織としての本来の生物学的特性を有するがん細胞集団を、インビトロで作製することを目的とする。 【解決手段】 本発明は、基板と生体適合性ポリマー層とを有する細胞培養基材であって、生体適合性ポリマー層で覆われていない所定の形状の所定の表面構造を持つ複数の粗面部分を所定の間隔で基材表面上に有する前記細胞培養基材に関する。本発明によると、所定の構造を有する細胞培養基材上でがん細胞を培養するという、とても簡便な操作により、生体内で観察されるものと同様の形態学的極性及び組織運動極性を有するがん細胞集合体を生きた状態で得ることができ、これにより従来不可能であったインビトロでの微小腫瘍のライブイメージングが可能となる。また、このがん細胞集合体は、生体内に生じるがんの発生、増殖、浸潤、転移、再発の一連の流れを再現していると考えられるため、がん研究におけるリサーチツールとして、又は抗がん剤のスクリーニングのために利用することができる。
