Methods to emulsify cells and/or mRNA with reverse transcriptase at a temperature such that the reverse transcriptase begins making cDNA during the emulsification. The disclosure provides methods for reverse transcribing mRNA into complementary DNA (cDNA) while simultaneously isolating cells into aqueous partitions. Methods of the disclosure provide for the very rapid capture of the information in mRNA into cDNA, which is more stable than mRNA. The cDNAs are made immediately as the sample is emulsified into droplets.

Procédés d'émulsification de cellules et/ou d’ARNm au moyen d’une transcriptase inverse à une température telle que la transcriptase inverse commence à produire de l’ADNc pendant l'émulsification. La divulgation concerne des procédés de transcription inverse d’ARNm en ADN complémentaire (ADNc) tout en isolant simultanément des cellules dans des séparations aqueuses. Les procédés de la divulgation permettent la capture très rapide des informations dans l'ARNm en ADNc, qui est plus stable que l'ARNm. Les ADNc sont produits immédiatement à mesure que l'échantillon est émulsifié en gouttelettes.
