Abstract
(EN) A linear double stranded deoxyribonucleic acid (dsDNA) molecule comprising operatively linked in the 5' to 3' direction: a) one or more Ter sites at the 5' terminus ("5' Ter"); b) a segment comprising a DNA sequence of interest; and c) one or more Ter sites at the 3' terminus ("3' Ter). Also methods of protecting DNA sequences of interest from exonuclease degradation using the DNA constructs of the present disclosure, cells transformed with the DNA construct of the present disclosure and cell-free synthetic biology system comprising a linear dsDNA molecule of the present disclosure.
(FR) L'invention concerne une molécule d'acide désoxyribonucléique bicaténaire (ADNds) linéaire comprenant de manière fonctionnelle et liés dans la direction 5' à 3' : a) un ou plusieurs sites Ter à l'extrémité 5' ("5'Ter") ; b) un segment comprenant une séquence d'ADN d'intérêt ; et c) un ou plusieurs sites Ter à l'extrémité 3' (" 3'Ter). L'invention concerne également des procédés de protection de séquences d'ADN d'intérêt contre une dégradation par exonucléase à l'aide des constructions d'ADN de la présente invention, des cellules transformées à l'aide de la construction d'ADN de la présente invention et un système de biologie synthétique acellulaire comprenant une molécule d'ADN bicaténaire linéaire de la présente invention.
