Abstract
(EN) A method for screening for a molecule capable of forming a complex in conjunction with a plurality of target molecules by applying a candidate molecule collection method utilizing an affinity technique in combination with a technique typified by a Split GFP technique is discovered. In this method, a first target molecule linked to a first segment in a protein, a second target molecule linked to a second segment in the protein, and a library including a plurality of test molecules are allowed to coexist. A test molecule which can form a complex in conjunction with the first target molecule linked to the first segment and the second target molecule linked to the second segment is collected by utilizing an affinity technique.
(FR) L'invention concerne un procédé de criblage d'une molécule susceptible de former un complexe en conjonction avec une pluralité de molécules cibles en appliquant un procédé de collecte de molécules candidates à l'aide d'une technique d'affinité en combinaison avec une technique caractérisée par une technique de protéine à fluorescence verte divisée. Dans ce procédé, une première molécule cible liée à un premier segment dans une protéine, une seconde molécule cible liée à un second segment dans la protéine, et une bibliothèque comprenant une pluralité de molécules de test sont autorisées à coexister. Une molécule de test qui peut former un complexe en conjonction avec la première molécule cible liée au premier segment et la seconde molécule cible liée au second segment est collectée en utilisant une technique d'affinité.
(JA) Split GFP技術に代表される技術にアフィニティーを利用した候補分子の回収方法を適用することによって、複数の標的分子と共に複合体を形成し得る分子をスクリーニングする方法が見出された。当該方法では、タンパク質の第1の部分に連結した第1の標的分子、該タンパク質の第2の部分に連結した第2の標的分子、および複数の被験分子を含むライブラリを共存させる。そして、該第1の部分に連結した第1の標的分子および該第2の部分に連結した第2の標的分子と複合体を形成することができた被験分子を、アフィニティー技術を使用して回収する。
