Abstract
(EN) The present disclosure relates to novel methods and compositions for a measuring exonuclease enzyme activity. In particular, the methods entail contacting a sample with a fluorescently-labeled substrate to create a test mixture, incubating the test mixture for a time sufficient for cleavage of the substrate, and measuring the fluorescence signal from the test reaction mixture. In certain aspects, the methods disclosed herein are useful for identifying and/or assessing a modulator of an exonuclease that may be used either alone or in combination with other compounds. The present disclosure also relates to fluorescently-labeled double- stranded nucleic acid compositions useful in the practice of such methods and, still further, to kits for performing the methods of the disclosure. The present disclosure further relates to compounds and methods useful for treating a viral infection.
(FR) La présente divulgation concerne de nouvelles méthodes et compositions pour mesurer une activité d'enzyme exonucléase. Plus particulièrement, les méthodes impliquent la mise en contact d'un échantillon avec un substrat marqué par fluorescence pour créer un mélange de test, l'incubation du mélange de test pendant une durée suffisante pour le clivage du substrat, et la mesure du signal de fluorescence émanant du mélange réactionnel de test. Selon certains aspects, les méthodes divulguées dans la description sont utiles pour l'identification et/ou l'évaluation d'un modulateur d'une exonucléase qui peut être utilisé seul ou en association avec d'autres composés. La présente divulgation concerne également des compositions d'acides nucléiques doubles brins marqués par fluorescence utiles dans l'exercice de telles méthodes et, en outre, des kits pour mettre en œuvre les méthodes selon la divulgation.
