Abstract
(EN) Disclosed are a nucleic acid isothermal amplification method and an application thereof. a reverse transcriptase, an RNA polymerase, and an RNase H used in the method are all heat-resistant enzymes still active at at least 45 °C, and an amplification reaction is performed at a constant temperature of not less than 45 °C. The isothermal nucleic acid amplification method of the present invention has the characteristics of being rapid, isothermal, highly sensitive, and highly specific, and is suitable for nucleic acid detection in a field such as clinical testing.
(FR) La présente invention concerne un procédé d'amplification isotherme d'acide nucléique et son application. Une transcriptase inverse, une ARN polymérase et une RNase H utilisées dans le procédé sont toutes des enzymes résistantes à la chaleur, encore actives à au moins 45 °C, et une réaction d'amplification est réalisée à une température constante non inférieure à 45 °C. Le procédé d'amplification isotherme de l'acide nucléique de la présente invention se distingue par sa rapidité, son caractère isotherme, sa grande sensibilité et sa grande spécificité, et convient à la détection de l'acide nucléique dans un domaine tel que les tests cliniques.
(ZH) 本发明公开了一种核酸的恒温扩增方法及其应用。该方法中所用的逆转录酶、RNA聚合酶以及RNA酶H均为至少在45℃仍具有活性的耐热酶,并且所述扩增反应在不低于45℃的温度下恒温进行。本发明的恒温核酸扩增方法具有快速、恒温、灵敏度和特异性高的特点,适合临床检验等领域的核酸检测。
