A number N-1 of metabolic chambers are provided on the upper surface of the lower chip layer, projections of the metabolic chambers on the upper chip layer respectively overlap with two upper fluid channels adjacent thereto, and each metabolic chamber is in communication with the two upper fluid channels adjacent thereto by means of the porous membrane layer. The porous membrane layer is made of a material which is impermeable to cells but permeable to cellular metabolic secretions, wherein N is an integer greater than or equal to 2. By means of providing the porous membrane layer allowing the transfer of metabolites, the metabolites generated by the cells and test drugs in the first upper fluid channels enter the metabolic chambers on the lower chip layer, and then the metabolites in the metabolic chambers enter the next fluid channel of the upper chip layer, the cells in the next fluid channel can continue to interact with the metabolites generated in the previous stage, and the influences of multi-stage drug-cell metabolism can thus be researched.
